Anti-CD30 Immunocytokine, scFv (HRS3)-IL12-IL2 (ACFP-SH175)

This antibody-cytokine fusion protein was achieved by conjugating/fusing the Anti-CD30 scFv to IL12 and IL2. It was expressed in CHO and purified with affinity chromatography. The Immunocytokines retains the ability to bind the CD30 as well as the biological activity of IL12 and IL2. The tumor-targeted IL12-IL2 fusion protein was superior in activating resting T cells to amplify and secrete pro-inflammatory cytokines compared to targeted IL2 or IL12 alone. This Immunocytokines was designed for treating Hodgkin's disease (HD) anaplastic large cell lymphoma.

Interleukin 12 (IL-12) is an interleukin that is naturally produced by dendritic cells, macrophages, neutrophils, and human B-lymphoblastoid cells (NC-37) in response to antigenic stimulation. IL-12 is composed of a bundle of four alpha helices. It is a heterodimeric cytokine encoded by two separate genes, IL-12A (p35) and IL-12B (p40). The active heterodimer (referred to as 'p70'), and a homodimer of p40 are formed following protein synthesis.
