The product specifically binds to the surface antigen differentiation cluster 4 protein. The antibody prevents infection of Tzm-bl cells by various subtypes of HIV strains, such as HIV NL 4-3 (subtype B), HIV 89.6 (subtype B), HIV Right G114 (subtype D), HIV Mj4 (subtype C), and HIV WCML249 (subtype D/C).

The HIV-1 isolate HIVNL4-3 (clade B) was analyzed for inhibition using the anti-CD4 monoclonal antibody 15A7 and its fragments using a luciferase-based virus neutralization assay in TZM-bl cells. The data are represented as the percent inhibition relative to the concentration of 15A7 and its fragments. The experiment was repeated four times to generate mean value and standard deviation.

The dot-ELISA assay for the serially truncated CD4 D1 domains that were displayed on HBV-C149 particles. Decreased concentrations of 15A7 were applied to determine the concentration-dependent reaction. HBcAg, CD4-D1 and CD4-D1D2 were used as controls.

Site-directed mutations of the gp120 binding sites were conducted on the CD4-D1D2 protein. The resultant proteins, F43A, K46A, R58A, R59A and N73A, were resolved in non-reducing and reducing SDS-PAGE and reacted with 15A7 via western blot. F43A, K46A, R59A and N73A lost binding capacity with 15A7, demonstrating that 15A7 recognizes a region that overlaps with the gp120 binding site on CD4.
