Recombinant Mouse Antibody (15A10) is capable of binding to Cocaine, expressed in Chinese Hamster Ovary cells (CHO). 15A10 hydrolyzes the benzoyl ester of cocaine to yield the nonpsychoactive metabolite ecgonine methylester with a kcat of 2. 3 min^-1, KM of 220 µM, and kcatkuncat^-1=2. 3 × 10^4.

The effects of 10, 30, and 100 mg/kg i.v. pretreatments of mAb 15A10 on operant responding for i.v. cocaine injections on a multiple-dose schedule are illustrated. Each point represents the mean (± S.E.M.) number of injections self-administered at a given dose of cocaine available within a single, 50-min component of the self-administration session (n = 4-5 per point). The average (± S.E.M.) of the three cocaine dose-response curves generated after saline pretreatments (n = 14 per point) in each of the treatment groups is also included for comparison.
