Anti-DNA Immunocytokine, IgG (chTNT-3)-IL12 (ACFP-SH139)

This antibody-cytokine fusion protein was achieved by conjugating/fusing the Anti-DNA IgG to IL12. It was expressed in CHO and purified with affinity chromatography. The immunocytokine retains the ability to bind the DNA as well as the biological activity of IL12. The activity of the expressed chTNT-3/huIL12 was confirmed by standard IL12 bioactivity assays which demonstrated that the fusion protein induced similar levels of peripheral blood lymphocyte (PBL) proliferation as free recombinant IL12. This immunocytokine was designed for treating tumor.

Interleukin 12 (IL-12) is an interleukin that is naturally produced by dendritic cells, macrophages, neutrophils, and human B-lymphoblastoid cells (NC-37) in response to antigenic stimulation. IL-12 is composed of a bundle of four alpha helices. It is a heterodimeric cytokine encoded by two separate genes, IL-12A (p35) and IL-12B (p40). The active heterodimer (referred to as 'p70'), and a homodimer of p40 are formed following protein synthesis.
