Anti-EDA Immunocytokine, IL12-scFv (F8)-scFv (F8) (ACFP-SH099)

This antibody-cytokine fusion protein was achieved by conjugating/fusing interleukin-12 as a single polypeptide with two F8 antibodies in single-chain Fv (scFv) format. It was expressed in CHO and purified with affinity chromatography. The immunocytokine retains the ability to bind the EDA as well as the biological activity of IL12. IL12-F8-F8 was capable of selective localization on the tumor neovasculature in vivo, as judged by quantitative biodistribution analysis with radioiodinated protein preparations. The IL12-F8-F8 mediated comparable patterns of leukocyte infiltration into the tumour mass and displayed a potent single-agent therapeutic activity. The protein potently inhibited tumor growth in three different immunocompetent syngeneic models of cancer. The treatment was generally well tolerated. This immunocytokine was designed for treating melanoma, lung adenocarcinoma, neoplastic or inflammatory disease.

Interleukin 12 (IL-12) is an interleukin that is naturally produced by dendritic cells, macrophages, neutrophils, and human B-lymphoblastoid cells (NC-37) in response to antigenic stimulation. IL-12 is composed of a bundle of four alpha helices. It is a heterodimeric cytokine encoded by two separate genes, IL-12A (p35) and IL-12B (p40). The active heterodimer (referred to as 'p70'), and a homodimer of p40 are formed following protein synthesis.
