Anti-EDA Immunocytokine, scFv (F8)-IL4 (ACFP-SH106)

This antibody-cytokine fusion protein was achieved by conjugating/fusing the Anti-EDA scFv to IL4. It was expressed in CHO and purified with affinity chromatography. The immunocytokine retains the ability to bind the EDA as well as the biological activity of IL4. F8-IL4 inhibited tumor growth in three different immunocompetent murine cancer models (F9 teratocarcinoma, CT26 colon carcinoma and A20 lymphoma). Furthermore, F8-IL4 showed synergistic effects when coadministered with immunocytokine based on IL2 and IL12. F8-IL4 was able to selectively localize to arthritic sites in vivo and exhibited a potent single-agent activity in the collagen-induced arthritis model in mice. This immunocytokine was designed for treating melanoma, lung adenocarcinoma, neoplastic or inflammatory disease.

The interleukin 4 (IL4) is a cytokine that induces differentiation of naive helper T cells (Th0 cells) to Th2 cells. Upon activation by IL-4, Th2 cells subsequently produce additional IL-4 in a positive feedback loop. The cell that initially produces IL-4, thus inducing Th0 differentiation, has not been identified, but recent studies suggest that basophils may be the effector cell. It is closely related and has functions similar to Interleukin 13.
