anti-ENG immunotoxin SN6 (IgG)-Ni-B (AGTO-L011N)
This immunotoxin was achieved by conjugating/fusing the Anti-ENG monoclonal antibody to Nigrin b. It was expressed in CHO and purified with affinity chromatography. The immunotoxin selectively binds to proliferating endothelial cells, then Nigrin b penetrates the cells by receptor-mediated endocytosis and depurinates mammalian ribosomes, thus inhibiting cell-free protein synthesis. It was designed for treatment of human solid tumors.

- Molecule Class
- L kappa; IgG1-Ni-B
- Toxin Description
- Nigrin b is a galactose-specific lectin isolated from the bark of elderberry, with two-chain structure and enzymatic activity similar to ricin. Both ricin and nigrin b depurinate mammalian ribosomes, a characteristic property of ribosome inactivating proteins (RIPs), thus inhibiting cell-free protein synthesis.
