Anti-EpCAM Immunocytokine, IL4-IgG (KS)-GM-CSF (ACFP-SH192)

This antibody-cytokine fusion protein was achieved by conjugating/fusing the Anti-EpCAM IgG to IL4 and GM-CSF, where one cytokine was fused at the carboxyl terminus of the H chain while the second cytokine was fused to the amino terminus of the light (L) chain variable region. It was expressed in CHO and purified with affinity chromatography. The Immunocytokines retains the ability to bind the EpCAM as well as the biological activity of IL4 and GM-CSF. These bifunctional molecules were shown to have striking anti-tumor activity as either gene therapy or as an antibody cytokine (s) fusion protein, and may provide a useful approach to the treatment of cancer. This Immunocytokines was designed for treating solid tumor.

Cytokines 1 Description
The interleukin 4 (IL4) is a cytokine that induces differentiation of naive helper T cells (Th0 cells) to Th2 cells. Upon activation by IL-4, Th2 cells subsequently produce additional IL-4 in a positive feedback loop. The cell that initially produces IL-4, thus inducing Th0 differentiation, has not been identified, but recent studies suggest that basophils may be the effector cell. It is closely related and has functions similar to Interleukin 13.

Granulocyte-macrophage colony-stimulating factor (GM-CSF), also known as colony stimulating factor 2 (CSF2), is a monomeric glycoprotein secreted by macrophages, T cells, mast cells, NK cells, endothelial cells and fibroblasts that functions as a cytokine. The pharmaceutical analogs of naturally occurring GM-CSF are called sargramostim and molgramostim.
