The neutralizing ability of each sdAb (10 lg/mL) was determined using 100 focus-forming units of JFH-1 particles before inoculating Huh7.5 target cells. Neutralization was compared with control neutralizing mAb 1:7 and an irrelevant antitetanus toxin sdAb.

Transmission between HCV JFH-1–infected Huh-7.5 cells and na€ıve Huh-7.5 cells in the presence of inhibitors was assessed via flow cytometry. Extracellular virus after 24-hour incubation was quantified via end-point titration, and a horizontal black line indicates the number of infected target cells in the presence of an antibody dose that eliminated>95% of cell-free virus. Inhibition of cell-to-cell transmission was defined as reduced infection below this threshold. D03 demonstrated dose-dependent inhibition of direct infection.
