Flow cytometry analysis showed dose-dependent binding of MIL5 to four HER2+ cell lines: SKOV3, N87, MDA-MB-175 and SKBR3.

MIL5 can inhibit 2C4_FITC with the IC₅₀ of 2.7 μg/ml (SKOV3) and 3.69 μg/ml (SKBR3).

Internalization of MIL5 in HER2-positive SKOV3 and SKBR3 cells. More antibody molecules entered SKOV3 cells (3 x 10⁶) than SKBR3 (1.2 x 10⁵), and MIL5 seemed to have weaker internalization function than 2C4 in SKBR3 cells.

ADCC detection of MIL5 in SKOV3 and SKBR3 cells on a dose-dependent manner. MIL5 induced a better ADCC effect than 2C4. A human anti-CD20 monoclonal antibody (TGLA) was set as isotype control (negative).
