Anti-ROS-CII Immunocytokine, scFv (1-11E)-IL10 (ACFP-SH004)

This antibody-cytokine fusion protein was achieved by conjugating/fusing the Anti-ROS-CII scFv to IL10. It was expressed in CHO and purified with affinity chromatography. The immunocytokine retains the ability to bind the ROS-CII as well as the biological activity of IL10. Viral interleukin-10 (vIL10) was fused to anti-ROS-CII scFv (1-11E) with a matrix-metalloproteinase (MMP) cleavable linker to create 1-11E/vIL10 fusion, 1-11E/vIL10 bound specifically to ROS-CII and to damaged arthritic cartilage. This immunocytokine was designed for treating arthritic joints.

Viral interleukin-10 has now been termed viral interleukin 10 (vlL-10) to reflect its protein sequence homology to "cellular" IL10 (cIL10, either murine or human IL10). It is now widely accepted that vlL-10 shares many functions with clL-10, principally, the ability to enhance survival of newly infected B cells and to diminish the production of IFN-β and IL-2 during ongoing immune reactions.
