Recombinant Human Anti-CHIKV E2 Antibody (CHK265) (CAT#: PABL-430)

Recombinant Human Antibody (CHK265) is capable of binding to CHIKV E2, expressed in HEK 293 cells. Expressed as the combination of a heavy chain (HC) containing VH from anti-CHIKV E2 mAb and CH1-3 region of human IgG1 and a light chain (LC) encoding VL from anti-CHIKV E2 proteins mAb and CL of human kappa light chain. Exists as a disulfide linked dimer of the HC and LC hetero-dimer under non-reducing condition. This antibody bound to CHIKV E2 B domain provided an explanation for its neutralizing activity. Binding to the B domain was associated with repositioning of the A domain of E2 that enabled cross-linking of neighboring spikes.

For immunoblotting, the protein was transferred from the gel onto a PVDF membrane and probed with commercial mAbs (Chk265), followed by incubation with HRP-conjugated anti-mouse IgG. Lane M indicates a protein marker (kDa).

As a spiking experiment, ELISA plates were coated with CHIKV-E2 protein (5 μg/mL) in PBS containing 1% human serum, blocked, and treated with the anti-CHIKV-E2 mAbs (10 μg/mL), followed by incubation with HRP-conjugated anti-mouse IgG. Commercial antiCHIKV-E2 mAbs (Chk265 and 16A12) were used for comparison.

After blocking with 5% skim milk in PBS, the plates were incubated with the Chk265 mAb (1 μg/mL), followed by incubation with HRP-conjugated anti-mouse IgG.
