Anti-EDB Antibody-Cytokine Fusion Protein, IgG-IL12 (CAT#: ACFP-SH086)
This antibody-cytokine fusion protein was achieved by conjugating/fusing the Anti-EDB IgG to the N-terminus of the IL12 p35 subunit, which in turn is disulfide-bonded to the p40 subunit.. It was expressed in CHO and purified with affinity chromatography. The immunocytokine retains the ability to bind the EDB as well as the biological activity of IL12. This protein has been shown to have a longer serum half-life than antibody-muIL12 in mice, and retains both antigen binding and IL12 activity in in vitro assays. This molecule also was found to have surprisingly low toxicity in immunocompetent mice. This immunocytokine was designed for treating Advanced solid tumors, Advanced pancreatic cancer, Metastatic melanoma.

- Interleukin 12 (IL-12) is an interleukin that is naturally produced by dendritic cells, macrophages, neutrophils, and human B-lymphoblastoid cells (NC-37) in response to antigenic stimulation. IL-12 is composed of a bundle of four alpha helices. It is a heterodimeric cytokine encoded by two separate genes, IL-12A (p35) and IL-12B (p40). The active heterodimer (referred to as 'p70'), and a homodimer of p40 are formed following protein synthesis.
