Anti-EDB Antibody-Cytokine Fusion Protein, IgG-IL12 (ACFP-SH087) (CAT#: ACFP-SH087)
This antibody-cytokine fusion protein was achieved by conjugating/fusing the Anti-EDB IgG to IL12. It was expressed in CHO and purified with affinity chromatography. The immunocytokine retains the ability to bind the EDB as well as the biological activity of IL12. It was found that huBC1-huIL12 has a longer serum half-life than antibody-muIL12 in mice. The antibody-huIL12 is a molecule designed to target tumor vasculature and evoke a localized immune cascade. The antibody-(IL12) demonstrated 10-fold lower IL12 activity compared with rmIL12 as determined by the level of IFN-γ secreted by PBMC. This immunocytokine was designed for treating Advanced solid tumors, Advanced pancreatic cancer, Metastatic melanoma.

- Interleukin 12 (IL-12) is an interleukin that is naturally produced by dendritic cells, macrophages, neutrophils, and human B-lymphoblastoid cells (NC-37) in response to antigenic stimulation. IL-12 is composed of a bundle of four alpha helices. It is a heterodimeric cytokine encoded by two separate genes, IL-12A (p35) and IL-12B (p40). The active heterodimer (referred to as 'p70'), and a homodimer of p40 are formed following protein synthesis.
