Anti-EDB Antibody-Cytokine Fusion Protein, p40-scFv (L19)-scFv (L19)-p35 (CAT#: ACFP-SH168)
This antibody-cytokine fusion protein was achieved by conjugating/fusing the Anti-EDB scFv to IL12. It was expressed in CHO and purified with affinity chromatography. The Immunocytokines retains the ability to bind the EDB as well as the biological activity of IL12. This p40-scFvL19/scFvL19-p35 retains IL12 biological activity, which was similar to that of recombinant murine IL12 (rmIL12) but without side effects. The p40-scFvL19/scFvL19-p35 also demonstrated potent anti-tumor activity. A single injection of this antibody-(IL12) fusion protein into mice bearing the F9 murine terato-carcinoma resulted in substantial growth retardation and decreased tumor volume. - Interleukin 12 (IL-12) is an interleukin that is naturally produced by dendritic cells, macrophages, neutrophils, and human B-lymphoblastoid cells (NC-37) in response to antigenic stimulation. IL-12 is composed of a bundle of four alpha helices. It is a heterodimeric cytokine encoded by two separate genes, IL-12A (p35) and IL-12B (p40). The active heterodimer (referred to as 'p70'), and a homodimer of p40 are formed following protein synthesis.
