Human Anti-EGFR Recombinant Antibody (clone Gc1118) (CAT#: PABL-080)

Recombinant Human Antibody (Gc1118) is capable of binding to EGFR, expressed in HEK 293 cells. Expressed as the combination of a heavy chain (HC) containing VH from anti-EGFR mAb and CH1-3 region of human IgG1 and a light chain (LC) encoding VL from anti-EGFR mAb and CL of human kappa light chain. Exists as a disulfide linked dimer of the HC and LC hetero-dimer under non-reducing condition. This antibody exhibited superior inhibitory activity against high-affinity EGFR ligands in terms of EGFR binding, triggering EGFR signaling, and proliferation compared with cetuximab and panitumumab.

HCT8 cell lines were pre-incubated with 0.1 μg/ml of cetuximab or GC1118 for 2 h prior to the addition of HB-EGF (20-500 ng/ml) or AREG (40-1000 ng/ml) for 10 min. Antibody binding to EGFR was assessed by measuring the fluorescence intensity using flow cytometry.

HCT8 cells were serum starved for 18 h, treated with 0.1 to 50 μg/ml GC1118 or cetuximab for 2 h, and stimulated with EGFR ligands (EGF, HB-EGF, BTC, and TGF-α, 250 ng/ml; AREG, 300 ng/ml; EREG, 500 ng/ml). pEGFR, pAKT, pERK, EGFR, AKT and ERK expression levels were analyzed by western blotting.

Athymic nu/nu mice were injected s.c. with HCT8, Lovo, HCT15, LS174T, LS513, and SW48 cells. GC1118 or cetuximab was administered intraperitoneally at 1 mg/mouse. N=6 animals/group. The data are plotted as the mean values±SD. Two-way ANOVA, **p<0.01, and ***p<0.001, compared with the PBS group.
