Mouse Anti-Fluorescein Recombinant Antibody (clone 43.1) (CAT#: PABL-096)

Recombinant Mouse Antibody (43.1) is capable of binding to fluorescein's carboxyl phenyl moiety, expressed in Chinese Hamster Ovary cells (CHO). It demonstrates a very high affinity (KD ∼ 70 pM) and a fast association rate (kon ∼ 2 × 10(7) M(-1) s(-1)).

Figure 2 Spectra and fluorescence decay of fluorescein, free and bound to mAb 43. Normalized absorption and emission spectra of free (A) and antibody bound (B) fluorescein. Excitation spectra fully trace absorption spectra in both cases. C: Fluorescence time decay of free and antibody bound fluorescein. To obtain comparable signal for the highly quenched bound form the sample concentration and excitation power were increased. D,E: residuals to the fit with three lifetime components.

Figure 3 Equilibrium binding plot and thermodynamic parameters of the mAb 43.1 - fluorescein interaction. A: The fluorescein concentration was kept at 20 nM while concentration of the antibody varied when performing equilibrium binding tittrations. Arrhenius plots calculated for the association (B) and dissociation (C) binding reactions and van‘t Hoff plot (D) with the Ka values calculated from the kinetics data.
