The initial reaction velocity (vi), normalised to the initial reaction velocity in the absence of Fab (vo), is plotted as a function of Fab concentration. The protease was held at constant concentration (50 nM). The curve shows the best ®t of the tight-binding model (Williams & Morrison, 1979) to the experimental data, from which the deduced value of Kinh is 35(3) nM.
