Recombinant humanized (from mouse) antibody expressed in CHO binding to human IgE Fc. Talizumab is a humanized monoclonal antibody as a new-concept therapeutic for allergic diseases. The unique anti-IgE antibody was designed to target immunoglobulin E (IgE) and IgE-expressing B lymphocytes specifically, without binding to IgE already bound by the high affinity IgE receptors on mast cells and basophils.

Western blot analysis of TAB-886 was performed by loading recombinant human IgE Fc protein in reduced condition Lane 1, Lane 2 and Lane 3 (0.1 μg, 0.3 μg and 0.6 μg respectively) onto a 12% Tris-HCl polyacrylamide gel. Proteins were transferred to a CN membrane and blocked with 5% skim milk for at least one hour. Membranes were probed with TAB-886 (2 μg/mL) and HRP-conjugated goat anti-Human IgG (H&L) as a secondary antibody (1: 5000). Chemiluminescent detection was performed.

Dot Blot analysis of TAB-886 was performed by coating with recombinant human IgE Fc protein.

ELISA analysis of TAB-886 was performed by coating with recombinant human IgE Fc protein. Then blocked with BSA and incubated with TAB-886. The HRP-conjugated goat anti-Human IgG as a secondary antibody (1: 5000). Detection was performed using TMB substrate and stopped with sulfuric acid. The absorbances were read on a spectrophotometer at 450 nm.
