Inclacumab (10 μg/mL) was added to anti-coagulated human blood prior to activation with TRAP (A), ADP (B), collagen (C) and AA (D). Maximal concentrations of TRAP (2 μM), ADP (5 μM), collagen (10 μg/mL) and AA (0.5mg/mL) resulted in ~70%, 40%, 10% and 20% P-selectinpositive platelets, respectively (data not shown). Values represent means ± SEM (n = 3).

Inclacumab was added to anti-coagulated human blood prior to activation with TRAP (25 μM). Activation of CD11b was detected on CD61+ CD45+-leukocytes using the CBRM1/5-PE mAb. Bars represent MFI of the CBRM1/5-PE binding expressed in percent of the TRAP-activated control (represented as 100%). Values represent means ± SEM (n = 3).

Human blood drawn from healthy subjects was perfused over collagencoated coverslips in the presence and absence of inclacumab. Each concentration was tested using blood from ≥4 human subjects (number of subjects is given on top of bars). Inhibition of leukocyte adhesion is given in % of the control adhesion per donor. Values represent means ± SEM.

The DrugMonitor™ Anti-Inclacumab Antibody (VS-1224-YC598) is an anti-drug antibody (ADA) against Inclacumab. This drug-based antibody is raised in mice immunized with the Inclacumab. The anti-Inclacumab antibody may be used in ELISA, pharmacokinetics (PK), and pharmacodynamics (PD) analyses, or serves as a reference standard in ADA assays. It also is an excellent tool for therapeutic drug monitoring, allowing to evaluate the drug efficacy and determine the drug concentration of the Inclacumab in samples.
