Recombinant monoclonal antibody to selectin P. Inclacumab (INN) is a humanized monoclonal that binds to selectin P.
Figure 1 Inhibition of TRAP-induced PLAs and PMAs in human blood.
Anti-coagulated human blood was spiked with the indicated inclacumab concentrations prior to TRAP activation. Inhibition of PLAs (A) or PMAs (B) is given in % of the TRAP-induced increase of PLAs or PMAs from non-activated baseline levels. Values represent means ± SEM (n = 3). The dotted line indicates the IC50 value.

Figure 2 Impact of inclacumab on PLAs induced by various platelet agonists.
Inclacumab (10 μg/mL) was added to anti-coagulated human blood prior to activation with TRAP (A), ADP (B), collagen (C) and AA (D). Maximal concentrations of TRAP (2 μM), ADP (5 μM), collagen (10 μg/mL) and AA (0.5mg/mL) resulted in ~70%, 40%, 10% and 20% P-selectinpositive platelets, respectively (data not shown). Values represent means ± SEM (n = 3).

Figure 3 Surface expression of the active form of CD11b on platelet-bound leukocytes.
Inclacumab was added to anti-coagulated human blood prior to activation with TRAP (25 μM). Activation of CD11b was detected on CD61+ CD45+-leukocytes using the CBRM1/5-PE mAb. Bars represent MFI of the CBRM1/5-PE binding expressed in percent of the TRAP-activated control (represented as 100%). Values represent means ± SEM (n = 3).

Figure 4 Inhibition of leukocyte adhesion to platelet monolayer in an ex vivo human flow system.
Human blood drawn from healthy subjects was perfused over collagencoated coverslips in the presence and absence of inclacumab. Each concentration was tested using blood from ≥4 human subjects (number of subjects is given on top of bars). Inhibition of leukocyte adhesion is given in % of the control adhesion per donor. Values represent means ± SEM.
