Antigen-specific binding of human IgG monoclonal antibodies was evaluated by direct enzyme-linked immunosorbent assay (ELISA). The heavy chain and light chain variable region (VH and VL, respectively) genes of the phage antibodies (a, D9; b, F6; and c, F10) were amplified and inserted into their respective human IgG expression vectors. Chinese hamster ovary cells (CHO) cells were transfected with these expression vectors and cultured for several weeks. ELISA was used to evaluate the antigen specificity of these antibodies in the culture supernatants.
