Recombinant Mouse Antibody (G17. 12) is capable of binding to PfMSP1-19, expressed in Chinese Hamster Ovary cells (CHO). G17. 12 antibody recognises a discontinuous epitope comprising 13 residues on the first epidermal growth factor (EGF)-like domain of PfMSP1-19.

This research presents a novel mass spectrometry-based technique called ITEM-THREE (Intact Transition Epitope Mapping-Targeted High-Energy Rupture of Extracted Epitopes) for mapping assembled epitopes on antigens. The study focuses on characterizing the interaction between a monoclonal anti-malaria antibody and the pfMSP119 antigen, an important protein from Plasmodium falciparum that remains attached to merozoite surfaces during red blood cell invasion. Using this approach, the researchers identified three epitope-carrying peptides from the pfMSP119 antigen that form an assembled (conformational) epitope. The findings demonstrate that ITEM-THREE can accurately identify complex epitopes with minimal sample consumption and relatively simple analysis procedures.
Creative Biolabs provided the monoclonal anti-pfMSP119 antibody (G17.12) that was crucial for this research. This antibody enabled the researchers to successfully map the epitope on the pfMSP119 antigen, contributing to the development of a potential screening assay platform for malaria. The antibody served as a positive control element that could help reduce false positive and false negative results in future malaria diagnostic assays, addressing a significant challenge in current rapid diagnostic tests which achieve sensitivities and specificities of only around 90%.
