Dead cells were excluded by FVS450 staining. OVA-specific CD4⁺ T cells were gated as CD4⁺ KJ1-26⁺ among live cells.

On day 14 after the transfer, CD4⁺ T cells were collected from the spleens of the recipients and adoptively transferred into wild-type mice. Wild-type recipients were simultaneously injected with OVA/IFA. Five days after readoptive transfer, draining lymph node cells were collected from the recipients and stainedwith anti-CD4 mAb and KJ1-26.
