A 20 μl volume of each sample was applied to one lane of SDS/PAGE and analyzed by immunoblotting with mAb H28-710. The cell lysate contained a large quantity of the inactive 13-kDa GIF that nonspecifically bound to Affi-Gel 10, but no 55-kDa GIF.

Aliquots of anti-CD3-treated F12 cells were cultured from 0 to 48 hr. Histidine-rich peptides in culture supernatants were purified using Ni-NTA agarose, and analyzed by SDS/PAGE, followed by immunoblotting with the mAb H28-710. The amount of each sample applied to one lane of SDS/PAGE was derived from 2 × 10⁵ cells.
