Recombinant Mouse Antibody (tAb2) is capable of binding to TGF Alpha, expressed in Chinese Hamster Ovary cells (CHO).

Cells (2 x 10⁴ were seeded in 0.3% agar containing DMEM and 10% FBS in 35-mm dishes and treated with different concentrations of mouse monoclonal anti-TGFa antibody TAbl or TAb2. The cells were grown for 14 d, stained with nitro blue tetrazolium, and counted with an Artek colony counter. (A) NOG-8 TF CI 5 cells; colonies greater than 150 pm for the untreated control cultures were 1022 colonies/dish. (B) NOG-8 TF CI 7 cells; colonies greater than 150 km for the untreated control cultures were 11 18 coloniesldish. Data are expressed as the percentage of cell growth as colonies in the presence of different concentrations of TAbl or TAb2 compared to untreated controls. Values are the average (± SD) of triplicate determinations.

Equal amounts of protein isolated from diierent ovarian tumours and non-malignant tissue as described above were resolved on an SDS-polyacrylamide gel and blotted onto a nitrocefhdose filter. TAB2 antibody was diluted 1:1000 and biotinylated rabbit anti-mouse and streptavidin peroxidase conjugate were employed to visualise immunospecilic bands.
