Mouse Anti-THPO Recombinant Antibody (clone TN1) (CAT#: PABZ-075)

Recombinant Mouse Antibody (TN1) is capable of binding to hTPO163, expressed in Chinese Hamster Ovary cells (CHO). hTPO163 interacts with soluble c-Mpl containing the extracellular cytokine receptor homology domains with 1:2 stoichiometry with the binding constants of 3. 3 × 109 M–1 and 1. 1 × 106 M–1.

rhTPO (14.1 fmol/ml) was preincubated with various concentrations of each Ab. FDCPhMpl5 (1 × 104 cells/well) were then incubated with these mixtures. After three days, the MTS assay was performed. Symbols represent anti-HT1 (●), TN1 (▲) and PCF9 (■). Results are expressed as the mean ± SD of triplicate determinations. The absorbance of the maximal cell growth (without Ab) and the background (without factors) was 1.06 ± 0.05 and 0.47 ± 0.01, respectively.

Three patients with acute myeloid leukemia in complete remission were treated with myelosuppressive chemotherapy. Serum TPO levels were measured by a sensitive ELISA (closed circles), and platelet counts were measured by an automated blood cell counter (open circles). The open triangles show the time of platelet transfusion (10 units). The arrows (↓) show the time of measurement of serum TPO levels and platelet counts just before and 1 h after platelet transfusion.
