Human Anti-ITGA4 Recombinant Antibody (clone HP1/2) (CAT#: PABL-360)

Recombinant Human Antibody (HP1/2) is capable of binding to α4 integrin, expressed in HEK 293 cells. Expressed as the combination of a heavy chain (HC) containing VH from anti-α4 integrin mAb and CH1-3 region of human IgG1 and a light chain (LC) encoding VL from anti-α4 integrin mAb and CL of human light chain. Exists as a disulfide linked dimer of the HC and LC hetero-dimer under non-reducing condition.

Wild type A4β7 and the indicated α4 mutant transfectants were incubated at 4 °C for 40 min with saturating concentrations of anti-α4β7 Act-1, anti-α4 HP1/2, anti-β1 TS2/16, or control P3 mAb. After washing, cells were incubated at 4 °C for 30 min with fluorescein isothiocyanate-conjugated secondary antibody (Dako, Glostrup, Denmark), and finally analyzed using a Coulter Epics XL flow cytometer.

Cells were labeled with 125I, solubilized, and the cell extracts immunoprecipitated with anti-α4β7 Act-1, anti-α4 HP1/2, anti-β1 TS2/16, or control P3 mAb. Immunoprecipitates were analyzed under non-reducing conditions by SDS-PAGE and autoradiography. The migration of the different forms of the α4 subunit and of the β1 and β7 subunits is indicated.
