Indirect eLISA results for detecting the binding of HuScFv in lysates of 19 representative huscfv-phagemid transformed E. coli clones to native NS1. Clones no. 11 and 13 (asterisks) were selected for further use as the HuScFv in lysates of these clones gave the highest eLISA signal to the antigen compared with the signal from background (HB) control.

The synthetic peptides of epitopes of HuScFv11 (DSGCVVSWKNKeLK) and HuScFv13 (TTNIWWSNVTA) could block the binding of the respective HuScFv to NS1 in the competitive eLISA in dose dependent manner while the irrelevant peptide control (WCDAFCSIrGKrV) did not affect the NS1 binding of HuScFv of both clones.
