ScFv antibodies were transferred to nitrocellulose membrane and probed with 9E10 antibody. Bound antibodies were detected using a HRP-conjugated goat anti-mouse and visualized with DAB.

Aggregation of A42 was monitored by dot-blot. Aliquots of samples were applied to nitrocellulose membrane (0.12 μg/dot) at the time indicated and then probed with 4G8 and scFv antibodies, respectively.

Figure 4 Effects of scFv antibodies on Aβ42-induced cytotoxicity.

Aliquots of Aβ42 preincubated with or without scFv's at indicated times were added to wells coated with SH-SY5Y cells. The final concentration of both Aβ42 and scFv was μlM. After an additional two-day incubation of cells and samples, the viability of the cells was measured using both MTT toxicity assay
