Rat Anti-ACVR1 Recombinant Antibody (clone 0443) (CAT#: VS-0724-YC41)
The product is a a rat anti-ALK2 antibody clone 0443. clone 0443 specifically bind mouse and human ALK2 among ALK1 through ALK7 with KD values of 5.1 and 5.6 nM, respectively. Clone 0443 is a blocking monoclonal antibody that can selectively suppress ALK2 signaling and prevents ligand-induced heterotopic ossification. Clone 0443 can induce dimerization of ALK2 extracellular domains in a back-to-back orientation on the cell membrane by binding the residues H64 and F63 on opposite faces of the ligand-binding site. Clone 0443 could prevent heterotopic ossification in a mouse model of fibrodysplasia ossificans progressiva that carries the human R206H pathogenic mutant.

- Function
- Bone morphogenetic protein (BMP) type I receptor that is involved in a wide variety of biological processes, including bone, heart, cartilage, nervous, and reproductive system development and regulation.
As a type I receptor, forms heterotetrameric receptor complexes with the type II receptors AMHR2, ACVR2A or ACVR2B.
Upon binding of ligands such as BMP7 or GDF2/BMP9 to the heteromeric complexes, type II receptors transphosphorylate ACVR1 intracellular domain.
In turn, ACVR1 kinase domain is activated and subsequently phosphorylates SMAD1/5/8 proteins that transduce the signal.
In addition to its role in mediating BMP pathway-specific signaling, suppresses TGFbeta/activin pathway signaling by interfering with the binding of activin to its type II receptor.
Besides canonical SMAD signaling, can activate non-canonical pathways such as p38 mitogen-activated protein kinases/MAPKs.
May promote the expression of HAMP, potentially via its interaction with BMP6.
