The anti-AFP pH-dependent antibody (VS-0825-YC212) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human AFP. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for AFP under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from AFP at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human Alpha-fetoprotein (AFP) is a major plasma protein produced by the fetus and is a well-known tumor marker in adults. Elevated levels of AFP are often indicative of hepatocellular carcinoma and germ cell tumors, making it a useful target for both diagnosis and therapy.
