The anti-CD160 pH-dependent antibody (clone CB52.6) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CD160. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD160 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD160 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD160 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CD160 is a glycosylphosphatidylinositol-anchored protein expressed on a subset of T-cells and natural killer (NK) cells. It functions as an inhibitory receptor, helping to regulate the activity of these immune cells, and has been implicated in chronic viral infections and tumor immunology.
