The anti-CD19 pH-dependent antibody (clone Taplitumomab) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CD19. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD19 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD19 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD19 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CD19 is a B-cell specific transmembrane protein that acts as a co-receptor for the B-cell receptor (BCR). It is essential for B-cell activation, proliferation, and differentiation, making it a primary target for B-cell depletion therapies.
