SmartAb™ Recombinant Anti-CD226 pH-dependent Antibody (Clone 10E5)

The anti-CD226 pH-dependent antibody (clone 10E5) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to mouse CD226. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD226 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD226 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD226 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Mouse CD226 is an activating receptor expressed on T-cells and NK cells that promotes cell-mediated cytotoxicity. It interacts with its ligands, CD112 and CD155, and is a key player in The anti-tumor and anti-viral immune responses.
