The anti-CD27 pH-dependent antibody (VS-0825-YC46) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CD27. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD27 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD27 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD27 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CD27 is a co-stimulatory molecule expressed on T-cells and is a member of the TNF receptor superfamily. Its interaction with CD70 promotes T-cell activation and survival, contributing to the development of long-term T-cell memory.
