  • The preparation method for antibody liposomes (P-Lipo) is as follows:
    1. Complex Formation: First, form the IRI-PC complex.
    2. Film Preparation: Mix the complex with a chloroform solution containing lecithin, cholesterol, DSPE-PEG2000, and DSPE-PEG2000-NHS in a weight ratio of 20/2/1/0.05. Subsequently, perform rotary evaporation to create a thin film.
    3. Film Hydration: Hydrate the thin film using a 5% glucose solution, followed by ultrasound treatment in a water bath.
    4. Extrusion: Subject the preparation to extrusion through polycarbonate membranes with pore sizes of 400 nm and 200 nm to obtain uniform liposomes.
    5. Removal of Unencapsulated Drug: Utilize a Sephadex G-50 column to remove any unencapsulated drug.
    6. Antibody Conjugation: Add anti-PD-L1 antibody to the liposomes, maintaining a molar ratio of antibody to DSPE-PEG2000-NHS of 1:100, and allow the reaction to proceed for 12 hours.
    7. Particle Size Analysis: Measure the particle size distribution of the liposomes using dynamic light scattering technology (Nano ZS 90).

  • The monoclonal antibody W2410F.9G2 specifically targets mouse PD-L1 (programmed death ligand 1), also known as B7-H1 or CD274. PD-L1 is a 40 kDa type I transmembrane protein that is part of the B7 family within the immunoglobulin superfamily. This protein is expressed on various immune and non-immune cells, including T lymphocytes, B lymphocytes, natural killer (NK) cells, dendritic cells, as well as monocytes stimulated by interferon-gamma, epithelial cells, and endothelial cells. The W2410F.9G2 antibody has been demonstrated to inhibit the binding of PD-L1 to both PD-1 and B7-1 (CD80).
