This mouse monoclonal antibody (mAb) is specific for PD-L1 and is designed to block its binding to PD-1 or CD80. The antibody exhibits a high affinity for PD-L1 at neutral pH but undergoes rapid dissociation from the antigen at the lower pH found within endosomes. The interaction with histidine residue 69 (H69) in PD-L1 is responsible for the pH-dependent binding properties of this antibody. Internalized mAbs are typically degraded within acidic endosomes. The rapid dissociation at lower pH facilitates antigen release within the endosome, which reduces the likelihood of lysosomal degradation and allows the antibody to be recycled for subsequent reuse. Human Programmed Death-Ligand 1 (PD-L1) is a protein expressed on the surface of various cells, including cancer cells, that acts to suppress the immune system. By binding to its receptor, PD-1, on T-cells, PD-L1 delivers an inhibitory signal that prevents the T-cells from attacking the cell, a mechanism frequently exploited by tumors to evade immune detection.
