The OKT3 antibody recognizes an extracellular conformational epitope on the ~20 kDa CD3ε subunit of the human T cell receptor (TCR)/CD3 complex. This complex is present on approximately 95% of mature T cells and NKT cells, and to a varying extent on thymocytes. Additionally, a majority of T cell neoplasms also express CD3. CD3 is a dimer composed of CD3γ, δ, ε, η, and ζ subunits, which are type 1 transmembrane proteins structurally related to the immunoglobulin superfamily. CD3 forms a non-covalent association with the TCR, playing a role in transmitting antigen recognition signals to the cytoplasm of T cells and regulating T cell activation and TCR expression on the cell surface.
