This murine anti-CEA single-chain variable fragment (scFv) antibody has been specifically isolated due to its high affinity for carcinoembryonic antigen (CEA), a marker often overexpressed in various cancers.   • Immunoliposomes were created by inserting scFv'-conjugated micelles into preformed PEG-liposomes. Liposomes made of EPC:Chol:mPEG2000-DSPE (6.75:3:0.25) containing 0.3 mol% DiI dye were prepared via film hydration-extrusion. Mal-PEG2000-DSPE micelles were formed by dissolving dried lipids in water and heat incubation. Reduced scFv were dialyzed to remove TCEP and mixed with micelles at a 5:1 molar ratio, followed by a 30-minute coupling reaction. Post-insertion of micelles occurred at specific temperatures for dual targeting with anti-EGFR and anti-CEA scFv', and unbound scFv were removed using gel filtration. Liposome size was measured, and micelles were stained with fluorescent labels for tracking during co-insertion.
