Human Anti-Citrullinated epitope Recombinant Antibody (clone C07) (CAT#: HPAB-AP043-YC)

Autoimmune reactions to certain epitopes of self antigens most likely contribute to the development of rheumatoid arthritis. Often these epitopes are citrullinated. Provided is a novel antibody C07 that can bind to certain citrullinated epitopes namely citrullinated enolase, vimentin, fibrinogen and citrullinated synthetic peptides. The antibody may also be an antibody that has reactivity against only CEP-1, CCP and cit-vim such as antibody C07. The antibody can be used in diagnostics of rheumatoid arthritis, for therapy against rheumatoid arthritis and as research tools.

Immunohistochemistry images showing brown 3,3-diaminobenzidene (DAB) staining of citrullinated targets in different stages of differentiation from CD14-positive monocyte precursors to Mφ and mature OCs. Slides were stained with murinised monoclonal anti-citrullinated protein/peptide antibodies (ACPAs) (mB02, mD10, mC07) and a monoclonal control antibody (mE02) and counterstained with haematoxylin (original magnification 500× for CD14-positive monocytes and mature OCs and 250× for the intermediate stages).

Applications

  • Application Notes
  • The Citrullinated epitopes antibody has been reported in applications of Immunohistochemistry, Immunofluorescence, Enzyme-linked Immunosorbent Assay.
For Immunohistochemistry: Cells were fixed with formaldehyde and stained with murinised monoclonal ACPAs (D10, B02, C07), murinised control antibody (E02), rabbit polyclonal anti-PAD2 and monoclonal mouse anti-PADI4 followed by horseradish peroxidase conjugated antimouse antibody as a secondary antibody and 3,3-diaminobenzidene (DAB). The slides were counterstained with Mayer's haematoxylin and viewed using a light microscope.
For Immunofluorescence: The cells were incubated with the murinised ACPAs, polyclonal ACPA and anti-CD68 monoclonal antibody, followed by Alexa-Fluor-633-labeled secondary antibodies (Abcam) and counterstained with 4′,6-diamidino-2-phenylindole.
