The anti-CLEC12A pH-dependent antibody (VS-0825-YC82) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CLEC12A. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CLEC12A under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CLEC12A at pH 6.0. This pH-sensitive antibody retains its binding affinity for CLEC12A on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CLEC12A, also known as MICL, is an inhibitory receptor expressed on myeloid cells, including dendritic cells and monocytes. It is a promising therapeutic target for acute myeloid leukemia (AML), where it is highly expressed on leukemic blasts.
