Mouse Anti-COL2A1 Recombinant Antibody (clone ACC1) (CAT#: PABC-248)

This product is a recombinant Mouse antibody that can recognize Col2a1. A mAb (ACC1) against citrullinated C1 was found to cross-react with several noncitrullinated epitopes on native CII, causing proteoglycan depletion of cartilage and severe arthritis in mice.

Reactivity of ACC1 towards denatured CI, CII, CIX, and CXI in ELISA. The Corning Costar plates (Thermo Fisher Scientific) were coated for 2 h with 5 μg/ml denatured collagen (70 °C, 30 min) and blocked with 3% heat-inactivated FBS in DPBS for 2 h at room temperature. Specific binding was detected using HRP-conjugated anti-mouse immunoglobulin kappa light chain antibody (clone 187.1; Southern Biotech) and ABTS (Roche) as substrate. Absorbance at 405 nm was measured with a Synergy-2 (BioTek Instruments).

Solid-phase inhibition assay. The ELISA plate coated by C1-CIT360 or C1-CIT365, was pre-incubated with ACC1 (2ug/ml) and then the sera (1:300 dilution) of a pool of 6 RA patients selected for positivity against C1-CIT360 was added for 3 hours of incubation at RT. The binding of human polyclonal antibodies to the given peptides was measured. One representative experiment was shown from three assays performed using triplicate technical replicates. Data represent mean + SD and the p value was calculated by Mann-Whitney U test to compare different groups.

    Inhib: A concentration 2 μg/mL of this antibody can be used for Inhibition.
