2.1.1D5 was identified from peripheral blood mononuclear cell (PBMC) samples in a longitudinal analysis of EVD patients. 2.1.1D5 binds EBOV GP but not Sudan virus (SUDV) or Bundibugyo virus (BDBV) GP. 2.1.1D5 was thought to bind the GP1 core region (outside the mucinlike domain) based on a competition assay and escape mutation analysis, in which Y232H was identified as a key escape mutation. Furthermore, the mutations G118E, T144A, and G224D blocked 2.1.1D5 binding to GP expressed on the surface of Jurkat cells (Jurkat-GP4).
