The anti-EGFL7 pH-dependent antibody (VS-0825-YC109) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to both human and mouse EGFL7. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for EGFL7 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from EGFL7 at pH 6.0. This pH-sensitive antibody retains its binding affinity for EGFL7 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. EGFL7, or Epidermal Growth Factor-like Domain 7, is a secreted protein that is involved in vascular development. It plays a role in the formation of blood vessels and is a potential therapeutic target in diseases like cancer.
