- Staphylococcus aureus PBP2a (Penicillin-Binding Protein 2a) is a cell wall synthesis protein unique to methicillin-resistant Staphylococcus aureus (MRSA)-a major cause of drug-resistant bacterial infections worldwide. Unlike other penicillin-binding proteins (PBPs) in Staphylococcus aureus, PBP2a has a low affinity for beta-lactam antibiotics (e.g., methicillin, penicillin). Normally, beta-lactam drugs bind to PBPs and inhibit their role in cross-linking peptidoglycan (the main component of bacterial cell walls), leading to cell wall collapse and bacterial death. However, PBP2a remains active even in the presence of these drugs, allowing MRSA to continue synthesizing a functional cell wall and survive treatment. The gene encoding PBP2a (mecA) is carried on a mobile genetic element called the staphylococcal cassette chromosome mec (SCCmec). Due to its central role in MRSA's antibiotic resistance, PBP2a is a critical target for diagnostic tests (e.g., PCR detection of mecA or immunological detection of PBP2a) and the development of new anti-MRSA drugs that specifically inhibit its activity.
