- Treponema pallidum p47 is identical to the aforementioned outer membrane protein of Treponema pallidum, the causative agent of syphilis. Its surface location makes it a key mediator of host-bacterial interactions, particularly adhesion to host cells-essential for Treponema pallidum to establish infection in the genital, oral, or anal mucosa. P47's strong immunogenicity is critical for syphilis diagnostics: detecting anti-p47 antibodies in blood samples helps confirm syphilis infection, especially in the secondary and latent stages. Research into syphilis vaccines has focused on p47 because targeting a surface protein involved in adhesion can directly block the bacterium's ability to colonize the host. Preclinical studies have shown that immunization with p47 can induce protective antibodies in animal models, reducing bacterial loads and preventing disease progression. Given the global burden of syphilis and the lack of an existing vaccine, p47 remains a top candidate for developing a preventive vaccine.
