- Treponema pallidum p47, as a major surface protein of Treponema pallidum, is essential for the bacterium's ability to infect and persist in the host. Its adhesion function allows Treponema pallidum to attach to host tissues, avoid being cleared by mucosal secretions, and initiate infection. The protein's immunogenicity makes it a reliable target for syphilis diagnostics, as anti-p47 antibodies are detectable in most infected individuals. Unlike some other Treponema pallidum proteins, p47 is not highly variable, meaning diagnostic tests targeting it have high specificity and sensitivity across strains. For vaccine development, p47's conservation and role in adhesion make it an ideal antigen: a vaccine could induce antibodies that bind to p47, preventing the bacterium from adhering to host cells and thus stopping infection before it starts. This makes p47 a critical molecule for both diagnosing and preventing syphilis.
