Recombinant Mouse Antibody clone Enro44, which is specific to Enrofloxacin. The antibody, Enro44, which can be used in a direct competitive ELISA to detect enrofloxacin with high specificity, sensitivity, and precision. It is sufficient for screening enrofloxacin in samples including chicken muscle, cattle milk, and urine within the range of the enforced MRL.

  • Enzyme-linked Immunosorbent Assay
    For hybridoma screening, MaxiSorp microtiter plates (Nunc, Roskilde, Denmark) were coated with 3 µg/ml of enrofloxacin-BSA in 0.1 M phosphate-buffered saline (PBS), pH 7.4, at 4°C, overnight. Plates were washed with PBS and blocked with 0.2% skim milk powder in PBS at 37°C for 1 h. After washing, the culture supernatant or sera were added, and plates were incubated at 37°C for 2 h. After another washing step, horseradish peroxidase (HRP)-conjugated anti-mouse rabbit antibody (Zymed, CA, USA) at a 1:2,000 dilution in 0.5% BSA was added to each well and incubated at 37°C for 1 h. The unbound antibody was removed by washing with PBS. Peroxidase substrate [150 µl of 0.01 M o-phenylendiamine dihydrochloride in 0.1 M citrate-phosphate buffer, pH 5.0, containing 0.001% (v/v) H₂O₂] was added. The reaction was allowed to proceed for 15 min at room temperature in the dark. The enzymatic reaction was terminated with 100 µl of 2.5 M H₂SO₄, and the absorbance was measured at 492 nm with a microtiter plate reader (Titertek Multiskan, CA, USA). For isotype determination, antibody isotyping kits (Sigma-Aldrich) were used.
