Antigens were immobilized in each well and incubated with various concentrations of antibodies. Bound antibody was detected using anti-human IgG conjugated to HRP. AH06 specifically bound to HER2, particularly its domain IV.

Antibody variants were injected at six different concentrations, 0.032 nM, 0.16 nM, 0.8 nM, 4 nM, 20 nM and 100 nM, over a surface on which 100 RU of HER2-ECD had been coupled. AH06 has a 7.4-fold improved affinity compared to that of the parent, hu4D5.
