The anti-F10 pH-dependent antibody (VS-0825-YC120) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human F10. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for F10 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from F10 at pH 6.0. This pH-sensitive antibody retains its binding affinity for F10 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human F10, or Coagulation Factor X, is a vitamin K-dependent serine protease that is a key component of the coagulation cascade. It is activated to Factor Xa, which converts prothrombin to thrombin, leading to the formation of a blood clot.
